Use of Model-Informed Drug Development to Streamline Development of Long-Acting Products: Can These Successes Be Translated to Long-Acting Hormonal Contraceptives?

چکیده
Long-acting contraceptives are the most effective reversible contraceptive methods. Increasing patients’ access to these may translate into fewer unintended pregnancies and lead substantial individual public health benefits. However, development of long-acting products can be complex challenging. This review provides ( a) an overview representative programs for antipsychotics as cases conceptual translation contraceptives, b) several case examples on how modeling simulation have been used streamline products, c) challenges andopportunities in developing information exposure-response relationships commonly progestins enable regulators developers rely prior findings effectiveness safety from approved contraceptives. The US Food Drug Administration is seeking assistance stakeholders provide data studies which pharmacokinetic pharmacodynamic or clinical outcomes hormonal were evaluated not previously submitted.

Long-acting reversible contraceptives (LARCs) are underused and many health professionals do not feel confident about how best to provide them. Research on improved treatment of HIV infection and pre-exposure prophylaxis (PrEP) continues. Poor adherence to treatment is the critical risk factor for virologic failure and resistance development, and long-acting formulations of anti-HIV medications that need only infrequent dosing may facilitate long-term therapeutic responses. However, such formulations and long-term persistence of drug a...

Volume 85, Number 4 www.aafp.org/afp American Family Physician 403 The use of long-acting reversible contraceptives (LARCs) has increased from 1.3 to 5.5 percent over the past decade. There are three LARCs available in the United States: the etonogestrel single-rod implant and two types of intrauterine devices (IUDs; copper T380A and levonorgestrel intrauterine system [Mirena]). متن کاملLONG-ACTING CONTRACEPTIVE AGENTS: TESTOSTERONE ESTERS OF CYCLOALKYLCARBOXYLIC ACIDS
The synthesis of twenty five esters of testosterone (17β -hydroxyandrost- 4-en-3-one) is described. All esters are derivatives of cycloalkycarboxylic acids. Some of the esters possess α- , β -and/or & - substitution in the ester side-chain. The work was undertaken in order to evaluate long-acting male antifertility effect of such esters. Most of the compounds, especially compound No.1, wer...
متن کاملUse of long-acting reversible contraceptives to reduce the rate of teen pregnancy.
Long-acting reversible contraceptives (LARCs) are safe for use in adolescents and do not rely on compliance or adherence for effectiveness. Continuation rates are higher and pregnancy rates are lower for adolescent users of LARCs compared with short-acting methods such as oral contraceptives. Similarly, repeat pregnancy rates are lower when LARCs are used compared with other forms of contracept...
